Kate is a sharpshooter and assassin who eliminates targets picked by Varrick, her loyal mentor and handler.Varrick reared her as a father figure after she was orphaned as a kid, giving her significant training in weaponry and warfare and finally inducting her into his own squad of wetwork experts.
As the film begins, Kate is in Osaka to assassinate an officer of a large yakuza syndicate, but Kate refuses to take the shot since he has been unexpectedly joined by a youngster. At Varrick's request, she eventually shoots the target.While Kate's task is a success, her violation of her own rule of not killing in the company of children causes her considerable distress. She informs Varrick that she will complete one more task before retiring to begin a new life. Kate encounters a fascinating man, Stephen, at her hotel's bar just before the last assignment. They have sex in her room and share a bottle of wine.
Later, while building her sniper's nest, she begins to experience disorientation, causing her to miss the shot. Kate knows Stephen poisoned her and, after wrecking her car, awakens in a hospital to learn she has acute radiation poisoning caused by Polonium-204 with just one day to live. She grabs injectable stimulant medication and a revolver and goes out to avenge the poisoner. Kate follows down Stephen and his girlfriend and discovers that Sato, a yakuza linked with the Kijima criminal family, coerced them into poisoning her. Kate tracks Sato down at a high-end restaurant and murders him with scores of armed yakuza. Desperate for information on the reclusive, well-protected boss Kijima, whom she suspects of being behind the poisoning, she kidnaps Kijima's niece, Ani. Kate discovers Ani is the girl who saw her father's death during the Osaka mission. While Kate first intends to use Ani as bait to draw Kijima out into the open, she eventually chooses to become her guardian after discovering that Ani's family intends to kill her as part of an internal power struggle sponsored by Kijima's unscrupulous adviser, Renji.
After hunting down Kijima, who explains that Renji made a deal with Varrick to merge his squad into the syndicate in exchange for killing Kijima and all of his blood relatives, Kate realizes that her faith in Varrick may be misplaced. Knowing Kate is on the verge of death, Kijima equips her with a small army of his soldiers to invade Renji's headquarters. Most of Renji and Varrick's men are killed in a gunfight, and Kijima personally beheads Renji for his treason. Kate tracks down Varrick, who is holding Ani hostage with a revolver. Varrick is fatally shot in the stomach as a result of the two's quick draw. Ani then assists Kate in walking outdoors to the roof, where she dies, finally at peace with herself.
